Hi, I did an ELISA Analysis for GAPDH. I followed the kit protocols and got OD values. I now also make a Stander curve. My question is that the units given in the kit are in ng/L. While enzyme activity is measured in U/L. I am totally confused about it. Can someone help me?
Example.
My OD value is 0.35
The stander curve equation is y=0.0471x-0.0064

U/mL is a mensure of enzyme activity. I am afraid your kit measures the quantity of the enzyme, but not its activity. If you want to quantify activity you have to set kinetic experiments.

You should talk with your supervisor before you collect any more data. You need to know what sort of data you are collecting and how you will be analyzing your data before you even start.
Iñigo Izal is correct that an ELISA cannot tell you enzyme activity, only enzyme concentration.
