I ran a batch culture and measured OD at 600 every half hour. I need to make a graph of growth rate /h. How to convert OD values to growth rate values at each point? I am attaching the graph I want to make for the reference.
Bacterial growth rate can be calculated from OD (optical density) using the following formula:
μ = (ln OD₂ - ln OD₁) / (t₂ - t₁)
Where: μ = bacterial growth rate (per hour or per minute) ln = natural logarithm OD₂ = the final optical density reading at time t₂ OD₁ = the initial optical density reading at time t₁ t₂ - t₁ = the time interval between the two readings
To calculate bacterial growth rate, follow these steps:
1. Take two OD readings of the bacterial culture at two different time points (t₁ and t₂), using a spectrophotometer or a similar device.
2. Calculate the natural logarithm of each OD reading (ln OD₁ and ln OD₂).
3. Calculate the time interval between the two readings (t₂ - t₁).
4. Substitute the values obtained in steps 2 and 3 into the formula above to calculate the bacterial growth rate (μ).
5. Express the growth rate in units per hour or per minute, depending on the time interval used in the calculation.
It's important to note that this formula assumes that bacterial growth follows a logarithmic (exponential) phase during the time interval between the two OD readings. If the growth curve deviates from this assumption, the calculated growth rate may not be accurate.
