I am using a Labomed Compound microscope to study the histology of layers of GIT and neurons. I have taken pictures on the magnification of 400X (10X eyepiece x 40X objective lens) and 1000X (10X eyepiece x 100X objective lens). How to calculate the scale bar for these images taken on these magnifications? How to add the scale bars in the images taken on this magnification?

You add scale bar on micrographs using image J software or Adobe Photoshop Elements™
Hi, you can add scale bar using ImageJ (Fiji) by;
1. open the image,
2. Draw a line (scale bar) on the drop it at your preferred location, say lower right corner, and
3. Use the line scale to determine the length of the line (scale bar) on the micrograph.
NB. You can adjust the length of the scale bar to give you a whole number such as 20 um or 10 um as the case may be.
I can show you how to do it for more clarity.
