I have always had trouble using square plates with media kept vertically. The sugar in the media leaks out in a few days even when thoroughly sealed with parafilm. due to which I am unable to study the morphology of seedlings. please suggest alternatives.
Why does the medium start leaking? Is it breaking down in sunlight, or does the plant root system metabolise it in some way?
Can you use a high-resolution photograph of the plants?
Can you place the plates at a 45degree angle to reduce leakage?
Can the agar content of the media be increased at all? Is there another gelling agent that you can use?
Do the plates have to be vertical?
Is there a different plate design available? It sounds like you are using a plate designed for horizontal work, when you really need more of a thin, flat but wide bottle design.
@Matthew wheal thankyou for the answer. I can increase agar content. We did try to place the plates at 45 degrees, but the leakage persists.
