I have to apply a Trizol protocol for the purification of RNA in viruses contained in culture media; in fact, it is the first time I use these techniques!!!!
Our goal is to obtain good RNA for metagenomics.
So anything you want to share with me, I'm all ears!!!!! :)
There are several protocols available (eg https://health.uconn.edu/li-lab/wp-content/uploads/sites/129/2017/06/total_rna_extraction.pdf)
However, if this is the first time you do it, I would strongly recommend you follow someone from your lab first who has experience with this. It is a fairly sensitive process, using highly toxic materials
The presence of contaminants such as inhibitors or RNases can affect the purity and yield of the RNA. It is recommended to follow good laboratory practices to minimize contamination and to perform quality control checks on the purified RNA.
@Rita I used Trizol RNA extraction from viruses and here is the protocol i used.
After 2 times washing of cells with cold dPBS and aspirate. add 500 microliters of Trizol per well ( 6 well plate) and incubate 10 min af rt shaker. wash well with Trizol sol. and add to a separate tube. (This can be stored in -80 celcius for later RNA isolation).
RNA isolation protocol is general.
