I am currently working in aptamer conjugated with gold nanoparticles. And for their conjugation thiol has to be employed. Some studies revealed that its being derieved from DTT but I am unable to understand that actual mechanism. Could anyone let me know how that thiol group is actually synthesized ?
It's not really derived from DTT. Usually the sulfur (which becomes the thiol) is added to the 3' or 5' end of the oligo during synthesis, most often as a disulfide. The role of DTT is to cleave the disulfide to generate the thiol terminus.
In some cases the oligo will actually have a thiol group when purchased. However, many people still treat with DTT before use and desalt, just in case the thiols have been partially oxidized.
