For miRNA delivery studies if conjugating and loading is required what temperatures and pH will it be stable at and what conditions should be avoided to prevent its degradation?
Synthesized miRNAs are usually get manufactured with slight chemical modifications to achieve higher stability, so if you store it at -20, and avoid frequent freeze-thaw then products remain stable for a year or more.
Now, the preparation of miRNA with transfection reagent and its delivery to the model, that much duration and room temperature condition does not affect the miRNA very much. So, once it gets transfected to the model, miRNA easily remains stable to carry out the function at cellular physiological conditions.
Degradation of miRNA won't be the first and immediate outcome at temp 30-40C. But it may depend on the synthesis of nanoparticle, and packaging process, if the process has a harsh treatment step then there is a chance of degradation. In my former lab, they packaged miRNA in dipeptide nanoparticle which consists of nanoparticle solution-miRNA vortexing and incubated at 37C for 30min. Further characterized it, found good and effective.
