Dear All,

I am real newbie in protein field. I would like to ask you a few question regarding to select band to measure in western blot.

In case that blocking is fine and I use single primary antibody (whether it is monoclonal/polyclonal) and detect multiple target protein bands more than it supposes to have according to the datasheet. How do I know what band is what? and What band I should use to measure?

I have tried to look info from article in pubmed on my target protein but most of them do not metion which kda band they use to measure. I also think the multiple bands may be its different isoforms, but How could I know which isoform do what? Also, Is it possible that the multiple bands can come from target protein binding to other protein and form different protein complexs? I am not sure about this because I always boil the extract with Laemmli sample buffer+BME before loading protein, so it should be denatured.

In addition, Could you kindly recommend me online protein database to look for protein info? espicially its signaling pathway, its form/isoform and its different function in each form.
