I plan to use Aβ25-35 to treat HT22 cells for establishing an Alzheimer’s disease model. Since oligomers are generally more toxic, they may be more suitable for cell modeling. I dissolved the peptide in triple-distilled water, which had already been autoclaved before use, so it should be sterile and safe for cells.

I have two questions:

  • In this case, does the prepared solution still need to be passed through a 0.22 μm filter?
  • To induce oligomerization, is there a recommended range for concentration, incubation time, and temperature? The methods reported in the literature seem to vary, so I would like to know if there are any official guidelines or best practices.
