Dear colleagues, how often should a cell culture be assayed during a 3 day experiment? Once at the start and then at the end or every 12 hours? We worry that every 12 hours is too stressful for the cells and may alter the experiment results. What's your opinion?

This depends on the aim of the experiment you do on cell culture
If you are experimenting with the stimulating effect of a substance on the proliferation of cells, I think it is possible to conduct the proliferation test every 24 hours or more .. because, and for a typical rapidly proliferating human, the cell with a total cycle time of 24 hours.
But if the aim is to study the toxicity of a substance on the cell proliferation ... the experiment can be conducted every 12 hours or even less

Here, I agree with the answer given by Mr Haddad. The assay time depends on the purpose of conducting the assay. If you are looking for any secondary metabolites such as enzymes, toxins or any other by-product produced by the cells. I think we should design the experiment targeting the time that product gives the maximum yield in the medium. If you are not quite sure about that particular time, usually what we are doing is we design a trial assay and withdraw samples from time to time and assay the particular product formed. Then we can find the time interval that we have to do the assay. Usually, that time depends on the proliferation time of the cell culture. It depends on the type of cell. Maybe 24 hours. However, if you are going to assay an effect of a toxin on cells e.g. susceptibility to antibiotics by cell culture assays time will be reduced; 9 hours.
