N.B: Our LnCap cells died after using 8 hrs starvation. We used 1000 cells/well. How can we get good results in MTT for LnCap cells? Your suggestions will be highly appreciated!
The period of serum starvation will depend on the cell type. It can vary from 1-6 hours up to 24 hours. In your case, you could try a pilot experiment where in you could subject your cells to varying periods of serum starvation and check the percent viability. From this experiment you would get an idea about the time point.
It is not necessary that you serum starve your cells for 24 hours before starting a cell-based assay. By starving your cells for longer periods, it could have a negative impact on your cells which in turn could affect your end result.
So, I would suggest you first optimize the length of serum starvation for LnCap cells before performing MTT assay.
