In this project that I have been given, we are to formulate a hypothesis and make a proposal as to how we would like to do experiments to test our hypothesis. One of the experiments that I will propose is gene induction using a tet-on system, but I have no idea how much time such an experiment requires and that needs to be included in the paper.
If one starts with an established cell line that allows the expression of a gene using a tet-on system, optimization of the induction of the gene and testing by real-time PCR and protein by immunoblotting (considering no technical issues) could take couple of weeks. If a cell system needs to be set up, it could take more than six months.
Additionally, if you are interested in a mouse model with tet-on system, it will take even longer. First, you need to either generate (with CRISPR, for example) a mutant with tet-operated expression of your gene of interest, or to purchase the existing mouse strain. It usually takes from 6 to 12 month to get fertile mutants (8 weeks old). Then you need to cross this mice with strain, containing tTA or rtTA transactivator. Such strains are usually available on market for certain cell types (expression of transactivator is linked to the expression of cell-type-specific gene/loci). It will take another 3 month to get fertile double mutants. So all in all it takes 1 year or so.
