I did the cloning and extracted the plasmids, now need to send the plasmids for sequencing. is it matter the concentration or no?

100ng/ul is ideal
To my view, when you are planning a sequencing, good to have 50-100ng/ul of DNA but this of course depends on the company used.
A rule of thumb is 100 ng of plasmid per kilobase pair (1000 bp) of total plasmid size.
