Dear all, I am trying to collect the conditioned medium of cancer cells after drug treatment and then add the conditioned medium to the HUVECs to see whether the tubes forms or not. How much serum is need to form the tubes? I found that tube formation fails at 1% serum and the results differ from high percentage of serum to low percentage of serum. I am wondering whether the final serum concentration would affect the tube formation. Thanks.

The addition of serum to the culture medium may influence tube formation behavior. In most cases, serum inhibits tube formation. To avoid this problem, test different serum concentrations, from 0 - 20%, with the conditions you define. This will help to determine which serum concentration is optimal for tube formation and cell survival.
