Hi, I have been trying to probe this blot with 6 antibodies, anti p-JNK, yap, p-Yap, Caspase9, Wg and tubulin. The first 4 are generated in Rabbit (cell signaling). Other than the obvious issue of air bubbles trapped during the transfer, can someone help me with the following issues.

1) All of these proteins are about 50KD but I get the exact 4 bands with each antibody. The probability of that happening seems low to me. Is there a way to confirm that the 4 bands I see are not artifacts?

2) No matter how much I wash I get all that background. I have tried 5% and 2.5% milk/ BSA, diluting primary and secondaries and increasing washing times.

3) What could be the possible reason for those vertical lines? I have never had them in the past.

The first lane on left is ladder. It should not light up but it does for some weird reason. The one next to it wild type and other lanes have experimental sample in different concentrations. The protein extract is from Drosophila larval tissue. I know that the stripping works as I have developed the blot after stripping and it is absolutely clean. Plus, the tubulin band looks neat without ant blotches or background and I see just one band for tubulin.
