How may I best identify primers when the sequence to be amplified is only 150 nucleotides downstream in the URR of a circular viral DNA?

This is for CRISPR-Cas9 editing of a conserved 16 bp segment with a 3' PAM sequence. The problem may be in the 5' primer selection.
