I have four DNA fragments for Gibson Assembly; two of the fragments are 5kb with a concentration of 0.011pmole/ul and 0.034pmole/ul, and the other two fragments are 0.5kb (0.163pmole/ul) and 0.6kb (0.236pmole/ul).

Based on the NEB suggestion, the total amount of DNA for the Gibson Assembly reaction is 0.2~1.0pmole with four DNA fragments.

Here is the question:

Should I consider the size of DNA to adjust the amount of DNAs I use, and if yes, how should I do the adjustment?
