It is almost two and half a months that I have started callus induction, co-cultivation and Agrobacterium mediated transformation in three different rice varities. Approximately three weeks ago I have transferred calli in the regeneration media (shooting media) and found that some calli regenerate but no shoot grows yet. Even still now I did not see any callus that turned greenish. I am confusing if I did any mistakes in the procedure! Is there any overall time table of grwoing shoot in the media? How long it may take to have shoot? Or after how many days I can confirm that there might be a problem in the process? I appreciate of having some suggestions in this regard. Thank you.
It's usually take time for regeneration of shoot after transformation, but I think it's better to transfer the greenish part of the callus to fresh medium with same hormonal combination and antibiotic included

The high concentration of hygromycin is lethal for plant growth and development. Also the genotype response vary with type of tissue culture media used and with concentration of hygromycin. Optimse your protocol using different concentrations (low to high) of hygromycin and for exposure time.
