Is there any part of you additional fragment that is fused with any of the packaging genes?
Does you pseudovirus has the viral dimensions to carrie this additional genetic information?
I would suggest a quick transfection for virus stock production followed by a series of Western Bolt to identify not only if you have virus production but also if you virus has all proteins that it is supposed to have.
In addition to that, take some of the supernatant from your virus stock production and use it to infect naive cells, in this way you can check if you have productive infectious pseudoviral particles.
Marcos, thank you for your advises ! My additional fragment do not fuse with the packaging genes .Now I'm trying to use the supernatant from the virus stock production to transduct 293T to see whether it will affects the efficiency of virus packaging .
