I am doing some splicing-related events of different tumour cells and it is supposed to starve cells before treating cells. One confusion is that how long should I starve the cells? Can I starve different cell lines for the same duration? Thanks.

It depends on the type of cell line, between 2 and 18 h is the most common. In my experience with chinese hamster ovary (CHO) cells, serum-starving for 16 h, before treatments, worked fine.

I agree with Maria. The duration of serum starvation will differ for different cell lines. Primary cells cannot tolerate serum starvation beyond 2 hours. The duration of serum starvation can range from 2 hours to 24 hours. So, to avoid any confusion, I suggest you perform a pilot experiment to get an approximate idea about the duration of starvation for different cell lines that you are using. You have to find out yourself.
You could expose your cell lines to starvation for different time points beginning with 2 hrs till 24 hrs. You need to add minimum quantity of serum, 0.2-0.5% during the starvation period just to maintain cell survival.
Observe the health status of cells at each time point. Do a cell viability check. If you are fortunate enough you could have a common time point for some cell lines, if not for all.
This experiment could help you to decide the duration of starvation for your cell lines.

To my best knowledge, different cell lines require different starvation duration, depending on the nature of the cells and the original tissue. I'd suggest you try to make a sort of titration or standardization. Thankfully, Malcolm Nobre had done that already with details.

sometimes it exceeds a week if you are working with cell lines. Overall, it depends on the tissue, organ and cell line used.
