Hi,
I have several samples to run RT-PCR, and wanted to know if I can prepare the plates in advance (Sample + Primers + SYBR Green), or if they need to be freshly prepared. Since I have so many samples, I thought maybe I can start preparing plates and then just reading them, without having to wait until the machine is ready to start a new plate.
Any ideas ?

Yes, You can Prepare plate in advance before run ended and put it in thermal cycler within hour.
If you do have thermal activation type of Taq you can prepare your SSR-type PCR several weeks earlier. I used Taq DNA polymerase such as ApliTaq gold which were inactive temperature below 50C.
