We typically culture cells for 24-72 hours to obtain a population of exosomes, but how quickly after the cells are placed in culture do they start spitting exosomes out? 1 hr? 6hr? Has anyone quantified a time course of exosome secretion?
exosome secretion takes place almost instantly, but as we typically want to obtian a decent number of exosomes- we let cells sit for at least 24h. use exosome-depleted FBS - from Gibco or obtained in house by ultracentrifugation - to ensure that cell derived exosomes are not contaminated by FBS exosomes (which are typically present in much higher numbers)
