I have to do SEM analysis of root samples colonized by a selected bacterium. After removal of roots from the pots, how long I can store the samples in 2.5% Glutaraldehyde solution. Is there any special treatment to increase the the storage period without affecting the colonized microbes?
Although I do not have enough experience experience in handling plant samples,I would however suggest to complete the examination under SEM as early as possible.
Rather than store glutaraldehyde, I would suggest that you continue the process and go through the dehydration series to the 70% ethanol stage, at which point you can store samples in a refrigerator. After that you can complete the dehydration and drying stages before EM work.

It should be safe. As Dilip Kumar Pal says it is best to continue with little delay. How long are you intending to store the material for?

If everything is fixed, rinsed, post-fixed, rinsed, and dehydrated through 30, 40, 50, 60, 70% alcohol you should be able to refrigerate and store. I have never stored anything that long, so I don't really know. It is usually an overnight store before continuation with dehydration.
Hello Kulandaivelu Velmourougane , I know that this post is almost 3 years ago.. but did you find a good method to store your plant samples before SEM analysis?
I have samples collected from the field and it is not possible for me have access to a SEM for a month. Thus, I am also looking for a method to store my samples for some time. Will be very useful if you have a method for it. Thanks!
