I want to prepare amyloid stock solution in DMSO. Article says sonication for 15 minutes at RT. they not mentioned intensity, I am confused, as this much long may be warm solution too much.
Hi Nirmal, 15 min sonication is way too long when using an ultrasonicator.
15 min in an ultrasonicator will heat and evaporate all the water and destroy the protein.
What they probably mean is 15 min in a sonicating waterbath. I have used sonicating waterbaths to solubilize amyloids and other proteins and it works great.
Thanks for answering. What is sonicating waterbath? i never listen about it so confused with it. Does amyloids are soluble in DMSO with out sonication?

Sonication of insoluble proteins pries apart big complexes and increases the surface area of the protein exposed to the solvent. Hence, faster solubilization.
