How long can I incubate my liver cells with BrdU?

I did a BrdU assay on the liver cells (L02) and the incubation time (with BrdU) was 24 hours. However, not much cells had the BrdU incorporated. Also, I do not have positive control. How do I troubleshoot? Will incubating the cells with BrdU for more than 24 hours be appropriate/harmful?
