I have worked a lot on isolation SRB, also using Posgate medium B. To be able to isolate a pure culture you need to do several practical things.
1) It is important to get "good" enrichment culture.
Good = You need to inoculate a vessel with medium by your sample and wait if bacteria starts to grow. Growth of SRB is indicated by formation of a lot of black precipitate! If you see so - transfer an aliquot (not a very big one) to another vessel and see if blackening is developed. ONly if you get a reproducible stable growth of SRB you may proceed further for isolation of a pure culture. Please, check your culture under the microscope - to see the type of dominating in numbers cells. Very often you may observe Desulfovibrio-like cells or small rods (Desulfomicrobium).
2) If (when) you get stable SRB enrichment culture you may try to isolate a pure culture using 10-fold dilution of culture in agarised Postgate B. A colony of SRB could be visible as a lense surrounded with blackening of the medium. However? you will succeed in isolate a pure culture only if in the inoculum used for isolation SRB cells were dominating over other non-SRB cells!!! That is why you need to get "enrichment" of the SRB cells in the "good" enrichment culture!!!

Perhaps, it is possible to use the protocol as for DNA extraction from soil.
