Currently, I am using CCK-8 to check cell viability on my desired assay plates. Can I, however, reuse the cells after using this reagent for additional experiments with this cell line?
Yes, you can reuse the cells after performing CCK-8 assay for additional experiments. This is because in CCK-8 assay, the water soluble tetrazolium salt,
WST-8 produces an orange formazan product upon bioreduction by cellular dehydrogenases which is soluble in tissue culture medium. The formazan product does not require any dissolution or solubilizing step as compared to MTT assay which requires a solubilizing solution to dissolve the formazan crystals.
WST-8 is not cell permeable as the electron mediator,
1-MethoxyPMS, receives electrons from viable cells and transfers the electron to WST-8 in the culture medium which results in low cytotoxicity.
So, compared to MTT assay, in CCK-8 assay, cells are least subjected to any kind of cytotoxicity. Therefore, you can reuse the cells for additional experiments.
