Actually I am going to transform my vector from E.coli JM 109 to Bacillus subtilis 168 I have completed my transformation and I need to get maximum OD600 by using different salts media. I already tried many salts media with different Nitrogen and Carbon sources but OD600 is too small. if someone has experienced about Bacillus subtilis 168 salt media and optimized growth please share relevant information about media and its conditions.

Try malt extract , rice malt or potato extract mixed with some essential salts as hit and trial attempt to have some initial inroads to later build upon it..
If you are using mineral salts and defined media you will not get the growth response of some undefined media based on organic not easily defined materials. I would like to suggest Bacillus subtilus grows like crazy on soybean based media. The bacteria is used to make a favorite Japanese dish of natto. If you imbibe soybean and autoclave them the inoculation of the beans will produce massive amounts of bacteria and by dilution you can get your target OD for a standard population for your use. The massive bacterial increase would best employ high temperature of 35 to 37 C and the increase should take just 1 to 2 days. Hope this useful for your studies.

Dear Muhammad what exactly is the defined growth media you are using. If you divulge I will give you some defined media options to test. But I do believe the soybean media would give huge amounts of bacteria to reach the OD requirement. Please share if you need a defined media and I will give some suggestions based on your defined mix.
