I am preparing a BSA solution for ChIP assays, the BSA solution will be used as a blocker for ChIP antibody binding beads. Should I prepare the BSA in Milli-Q water or PBS or TBS or is there any specific buffer that I should use to dissolve the BSA powder? Thanks!
I usually make a 3-5% BSA stock (Molecular Biology grade) in 1X autoclaved PBS (which prevents DNase from remaining active). and use this as blocking agent for my ChIP magnetic beads. During blocking and antibody binding I also add protease inhibitors and the binding reaction can proceed for upto 1hr at Room Temp or upto 4hrs at 4degC.
Thanks Subhojit! Wonder if I can use my dilution buffer to dissolve the BSA (which is not PBS or TBS)?
If manufacture given ChIP Dilution buffer contains detergents it will not allow full blocking of the beads. So i would be cautious.
