If you are having trouble measuring the protein concentration using the BCA assay, two possible reasons are (1) the protein is too dilute to detect (less than about 0.3 mg/ml) or (2) there is a substance in your sample that is not compatible with the assay. Check the BCA product literature. If the problem is incompatibility, try using either the Lowry assay or the Bradford assay, which have similar sensitivities as the BCA assay but may not have the same incompatibility issue.
