How does a single H3 mutation capable of affecting the global level of H3K27me3?

In DIPG patients, one puzzle is that typically only one copy of the H3 gene is mutated. Therefore, how does one single copy of mutation affect the global level of H3K27me3, considering that there are many H3 copies in the genome?
