I am working with magnetic nanoparticle coated/capped with different plant extract. I want to perform DLS analysis. Can anyone help me in preparation of sample for DLS. How much nanoparticle should I dissolved in water? Also will the amount of nanoparticle affect my reading?

For efficient scattering of light, it is always advised to keep the solute content as less as possible. Even 1% or less of your solute dispersed in proper solvent can provide you better results.
For Magnetic nanoparticles kerosene is a very good dispersion medium, but as you mentioned that your nanoparticles are coated with plant extract i am not sure whether that works in your case or not. If you can achieve a stable dispersion in water its the best for you or else you can go for solvents that doesn't effects the nature of your coated nanopartilces.

Another good way to decide a proper solvent (dispersing medium) is the intended use of the particles. If it will be mostly applied in aqueous media then you should really consider dispersing them in water.
Second, ultrasonication is important as Kamyer mentioned. I suggest you used bath sonication. I have seen people sonicate for as low as 20 min and as high as several hours. You might want to try a couple of different parameters so how how your particles respond.
pH of water may be adjusted slightly by your particles so it is a good idea to determine pH after introducing them. If necessary you might want to adjust pH with acid/base or a buffer, depending on intended application. I always prefer to measure hydrodynamic size in low ppm range (e.g. 10 mg/L).
