what is your substrate ? if your substrate is transparent then u can measure absorption through transmitting mode simply..if your substrate is opaque, then you have to measure with reflection mode..it will give you overall effect of both combined silver and CdSe abs spectra...else you can do the background subtraction with both silver film at first (auto zero)... this serves the minimization of silver effect and will give you pure CdSe QDs abs spectra separately....
Do you want to measure single CdSe QD spectra, or an ensemble spectra ?
The second option should be "easy" to implement and a UV-VIS plug-n-play device should do the job.
The first option is more of a tuff job and requires state of the art single molecule techniques like differential transmission (Check the work of Sandoghdar) or photothermal measurement (Check Lounis work). In both cases, you have to build (or buy) a confocal microscope.
