How do I solubilize protein without denaturing it and without using detergent or Urea?

I have a Brugia protein. It is getting expressed in the pellet. I need to solubilize it in a way that it does not lose its activity and its should not get denatured in the process. I cannot even use detergents as I fear it might alter my biological assay. Can anyone suggest me a technique?

Hi Amruta,

if your protein is in a pellet it has precipitated and has lost its functionality already. Perhaps you can re-dissolve the protein in urea or guanidine-HCl and refold it by slowly removing the denaturing agents (for example with dialysis).
