hello. In our lab, we incubate e.coli for 16 hours at 37 degrees Celsius and 130 rpm. We then streak and incubate at 37 degrees Celsius and 225 rpm and process IPTG when the O.D. reaches 0.6. I thought I hadn't changed anything from my usual routine, but the e.coli is growing slower these days. The O.D value used to reach 0.6 in as little as 2 hours after streaking, but now it takes longer than 2 hours to reach 0.6. How do I solve this problem ?

PS. Will fixing the worn probe of the sonicator solve the problem of under-picked proteins ?
