I'm currently working on the proposal for my first project. the plan is to take the samples with swabs, place them in TSB then use the bacteria from the broth in a cell culture. how do i quantify the amount of bacteria in each broth? the method I've seen is to make a curve using known suspensions but does that work for a complex culture? i was thinking maybe using a mock population of the microbiome to make a curve will fix the issue

Hello if spectral method not working, better to go to classical method with decimal dilutions, no other method which give you more reliable picture than this.

I agree with Karen's method of counting bacteria.I suppose if you want to get an overal count of bacterial cells growing in that medium whether it is complex or not, all the culturable bactrial cells will be included in your colony count when you count colonies appeared on plates.If you know the volumes of inital broths you plated then you can back calculate the number of cells present in your sample.By the way, I suppose it is always will be a gross calculation, not an exact value.
