I'd like to perform confocal Z-stacks on a non-adherent/suspension cell line, following different nanoparticle treatments.
How can I best 'restrain' these cells, so that they remain stationary during the Z-stack imaging?

There are several methods to immobilize cells in suspension. My favorite method is freezing in OCT after fixing and performing a sucrose gradient.
Another option is to place a tiny drop of fixed cells on a slide and immediately mount a cover-slip and proceed with imaging. Sealing the edge of the cover-slip is useful for longer imaging sessions.
