I want to develop growth curves for three different Lactobacillus species. They all seem to grow fairly well on MRS media at 35 C and 5% CO2 but not that well in MRS broth under these same conditions. I am concerned about how long and in what volume to make my started culture before it would be ready to start monitoring growth and doing plate counts? Any advice appreciated. When I try to look this up I get a lot of conflicting advice
Instead of using CO2 you can use nitrogen for producing lactobacillus in broth. You will get good growth
thanks for the suggestions but what I need to know is if I take a colony and innoculate a 100ml broth sample ON, then do a 1:10 dilution and read the OD 600 and it indicates that its already in log phase, do I then dilute that again to start the growth curve so I can watch the doubling time?
