i did not encapsulate any thing yet, but i assume that if i have encapsulated peptide in cyclodextrine and i would like to extract the peptide from these capsules., what i have to do?
As I mentioned, by changing the buffer conditions. It'll help you to alter the hydrophobic interactions or to disturb the peptide tertiary structure.
Compounds complexed by cyclodextrins are released upon water contact.
So, if you want to extract peptides which are complexed by cyclodextrins, you should extract the complex with water or aqueous buffers (e.g. isotonic PBS buffer). You should follow the peptide release over time to find optimum release. The release could be followed either by UV spectroscopy (if your peptide contains aromatic amino acids), by ninhydrin reaction (reaction with free amino groups), HPLC analysis or other methods.
