There is often a variations of concentration of PGR required for the in vitro culture of plants. Some plants may require very low concentration whereas other may require higher concentration. So is the any method to determine the concentration of the same depending upon the plant type.
Whether you are going to use PGRs in your culture medium, which ones and in what concentration, depends on:
1) What plant species you are growing and which type of explant: and
2) Which physiological process you are trying to induce (germination, organogenesis, somatic embryogenesis, micropropagation etc).
Your first trial should be to grow plants on a nutrition medium without PGRs as the use of PGRs may induce somaclonal variation (see the paper of Neelakandan and Wang, attached). Murashige and Skoog (MS) medium is used successfully for most plant species, but you might want to try some other media, such as WPM (woody plant medium) if you're growing a woody species, and so on.
If you're not obtaining satisfactory results with a medium without PGRs, and you've made sure that the media composition, pH and other parameters were correct, only then you should start looking for application of PGRs.
Depending on the plant species and type of physiological process, try first to find out which protocols have been used by other researchers who have done the same thing that you want to do, in the same species or at least in a related species (such as, species of the same genus or family). Try reproducing their protocol, then try optimizing it by varying the concentrations of PGRs.

Dear Gaurab, I fully agree with Martin. In addition i would say that the Type and concentrations of PGR is highly dependent on so many factors including but not limited to the endogenous PGR levels in the explant, age and stage of explant, physiological state of the explant (Actively growing or dormant), soft, semi soft, semi hard wood and hard wood (nature of explant), explant from current year growth or last year growth, juvenile, young or mature explant, apical or lateral bud, internodal or nodal, leaf or bud to name a few. Even with in same plant if you try different explants, they will all have different responses to same PGR as well as similar responses to the different PGRs. Apart from these factors that may affect the responses, the purity levels of media components, agar as well as PGRs are also one of the very big source of variation, as even minor elemental contamination can have profound impact/adverse response in the plant tissue culture. As Martin suggested, try reproducing the protocol of other researchers as a first step. Hope this helps to clear some of the doubts you are facing. Good luck.
I suggest to carry out dose-response curves for your plant cultures. There is not an equation, or mathematical function, to achieve those values for each plant specie. Otherwise, trial and error to determine the proper concentration of PGRs to determine an effect over your plant culture.

I suggest the same trial and error method if you want callus go for more auxin 2-4mg/l less cytokinins 0.25-1mg/l the reverse for shoot induction
