For staining method, either way is used not only for dying the microbes by destroying the cell wall structure so that the dye solution can penetrate into cell, but actually for adhering the cell on the plate not to wash away from any solution. Here, my question is the precise interaction between cell and plate. What makes these two objects strongly binding each other after fixation?
My first guess is that molecular interactions between silica of glass plate and macromolecules of cell membrane, such as hydrogen bonds and hydrophobic interacton are increased by giving the cell into coupled amount of surface dimension.
Additionally, both heat fixation and alcohol fixation are used for sample with agar. Then, would agar compartment be like the sticky frame, which helps cell binding to the glass plate?
I have one solution. You can try use a Fisher ProbeOn Slides. I use for in situ hybridization. In this procedure I wash the slides in alcohol series. Ande the section are keep adhere to the slide.
It happens becauae the slides is positivecharged.

It's more or less the same thing that happens when you cook an egg in a skillet. The heat causes the protein to denature which exposes a large number of charged moieties that were previously hidden or masked inside the protein. This makes egg protein or bacterial protein very sticky. It will then attach by electrostatic interactions to other charged material like glass which is highly negatively charged. This denaturation also occurs when proteins are exposed to high concentrations of alcohol.
Yours is a very fundamental question. Do a few small experiments; take very small amount of dextrose on a glass slide and heat it. See it it sticks on it after cooling. Ask yourself why did it happen? Similarly take a small amount of peptone on a glass slide and do the same experiment and find out why it sticks on after heating and cooling. Repeat the experiment with magnesium sulfate and ask the same question. Now repeat all the experiment with alcohol instead of heating and ask the same question. Of course keep in mind you basic chemistry knowledge to get the answers.
