I have tried to break DNA by water sonicator, but after gel run i saw that nothing happen. I took 300 μl DNA in eppendrop and flaot it on the bath, temp was 28-34`C, i sonicate the sample in three steps each 4 min apart, total 12 min.
would you please give me some suggestion?
Hi Asim Haque . If you don't have a tip sonicator, you can shear DNA by pushing it through a small bore syringe needle.
