You do not need COSY if your signals are unequivocal. Cis isomer has smaller constant coupling tan trans congener.
If you are not restricted to a COSY and if your are investigating peptides or proteins, just measure a 1H-1H TOCSY. The cis and trans isomers conected to Xaa-Pro isomerism are easy to identify.
