To screen an extract what type of cell lines is preferred? Is there any preliminary test to do?
You should began to test your extract for normal proliferaive cells like lymphocytes !

I have a doubt that,to screen for anti cancer property on what type of cell line like KB(oral), NCL-H46(lung), MCF-7,U-87, HeLa, NCL H-46 can we test for activity.
I think we use normal lymphocytes for testing genotoxicity by SCGE.

For Anti cancer screening to you extracts, you can find attached reference link (nature review) which has been developed by the US National Cancer Institute (NCI) 60, whom developed a tumour panel for anticancer drug screening. Cell lines were assembled that ultimately represented nine distinct tumour types: leukaemia, colon, lung, CNS, renal, melanoma, ovarian, breast and prostate.

Also has Ridha Oueslati said primary lymphocytes can be used for initial screening to evaluate cytotoxicity; And they are also useful for screening Genotoxicity studies like chromosomal abberation (OECD 473) and Micro nucleus test (OECD 487); which reveals the initial site of action

We use the following cell lines for preliminary screening of anti-cancer compounds namely, HCT-116 (Colon) , NCIH 460 (Lung) and U251 (Neural). Try to follow the method of NCI anti-cnacer drug screening protocol for the determination of In vitro toxicity ( using GI50, TGI and LD50).
