In order to determine the function of macrophage subtypes, we measure the amount of cytokines they produce by ELISA. However, we add recombinant IL-10 to induce the subtype we want. This makes measuring the IL-10 produced by these macrophages quite difficult, as ELISA is unable to distinguish between endogenous and recombinant IL-10. Does anyone have advice on how we can distinguish between these two types of IL-10?
