I am working with gelatin/PCL core-shell nanofiber fabrication, in this case I need to crosslink my fiber sheet then use it. I chose 25%(w/v) glutaraldehyde to use its vapor crosslink the nanofiber for 1 h. But my professor asked me how to know the nanofiber sheet crosslinked is sufficient, and he also said there is some ways to know how to measure the directly value of crosslinked efficiency. Can someone help me in this way? Thank you so much.
As Amin says, what would be sufficient for your experiment question.
If you focus only on Gelatin: You could do extraction test with water after different times to see how much gelatin isn't fully cross linked.

1. Cross linking will directly increases the mechanical properties, thus by measuring the tensile strength using Universal Testing Machine(UTM) you can justify the effect of cross linking.
2. Sufficiency of cross linking depends on the application of our use, thus by varying the parameters like percentage of cross linking agents used, temperature, time, etc and by corresponding measure of mechanical properties will helps you to find out the sufficiency of cross linking.

Regarding your description you are trying crosslinking gelatin that is inside of PCL fibers with GA vapor, meaning that you only crosslinking the end of fibers, right? I guess FT-IR is much faster when you check the presence of amino group peak. The more crosslinking the less intensity of NH peak.
