I have been working with phospholipid vesicles and encapsulation of bio-molecules or drugs. But, i am unable to ensure whether it is taking place or not. what are the methods to do so including UV-Vis, PL. For example, if a polymer or a drug (dox) is my target to attach, how can you go for it with confirmation. Then, if it happens, can we quantify how much is encapsulated. Is there any characterization tool which can help in visualization of the system that is actually occurring.

There are many ways to separate the encapsulated substance from the one that's in the surrounding aqueous media. They depend on the nanocarrier and the encapsulated drug properties and include cetrifuge filtration, vesicle dissolution followed by HPLC, dialysis techniques, and maybe others.
To find out in detail, review a drug delivery paper that discloses such parameters as encapsulation efficiency and loading capacity of the nanocarrier. In methods, they usually describe the separation of the encapsulated drug from the free drug.
Rais Pavlov thank you for the answer. can you please explain how to monitor dox loading into the vesicles using particularly UV-Vis spectra. Actually, for separation of free drug, i normally use centrifuge and dialysis technique. but, i am not sure whether i am able separate them or not from the loaded one.
After encapsulation, transfer the system into new vessel and dissemble the liposomes in some solvent. Now, analyse the soln in the UV and compare with the concentration curve.

Omkar S. Kushwaha how do you explain encapsulation in terms of uv-vis spectra.or how can encapsulated liposome be identified from uncapsulated one. then, for separation, how can we proceede.
