I need to know the detailed procedure for verification of a calibrated ELISA reader.

I do use a calibaration plate form our vendor (http://www.biotek.com/products/accessories/absorbance-test-plate/) But may be you can also use serial dilutions of a dye and lamda scans to verify your instrument.

The procrdure might differ depending on you device. We are using a BioTek Synergy HT. The light for the absorbtion measurement is generated by a monochromator. So I can preform a lamda scan with i.e. every 5 nm a measurement (by software). The resulting curve of i.e. a dye like Trypan blue should be resamble the known spectra specialy with in the peak areas.
I you know take the dye and preform a serial dilution (in doubles) the linearity at the maximum absorbtion wave lenth, should be given at least for absorbens between 1-0.05.
If you have a calibration plate or Filter with a know optical density at wave lenth (xy) you can also use that for the verification.

Will this "calibration plate" have traceability certification along with it?

at leat our calibration plate, came with a certificate which shows the manufactures data for each density filter and wave length and it comes with a date of expire or recalibration by the manufacture.

My chemistry involves the usage of 550 nm filter/wavelength. Can you suggest any stable dye/solution to check the ELISA reader linearity/calibration?
