I need to verify whether my treatment may alter the invasive properties of a human neuroblastoma cell line. I have a PerkinElmer Victor 3 microplate reader. Does anyone know a reliable and widely-accepted method? I have found some invasion assay kits (CellBioLabs and Millipore) but they really cost too much! Moreover, I do not have the 560nm filter which is required by the kits. I could also prepare invasion chambers by myself but I could really use some help to save time and money. Thank you very much in advance.
Typically, you would use invasion chambers coated with matrigel. As such, you don't really need a kit. Just get the invasion chamber, some matrigel, and some crystal violet to stain the bottom of the chamber. You can the quantify using Image J, a free available software.
96 well plates need to be coated with matrigel. and u need crystal violet. tht would do.
Type I collagen is also useful as Matrigel. Depending on the cell context, the stiffness of ECM microenvironment should be optimized. In the case of neuroblastoma, the stroma is not so stiff as that of pancreatic cancer or signet gastric cancer, which is why matrigel or the combination with type I collagen would be appropriate.
