As I know, Jurket E6.1 is naive T cell, so we need to stimulate them to become CD8 or CD4T cell.
Does anyone know which stimulator I should used to differentiate Jurkat E6.1 cell to become effector CD8 T cell, or CD4 T cell?
I am a master student and I felt very lost about this. The previous person treated this cell with PMA to produce IL-6, IL-10. If want to see TNF-a, treating with PHA. She does not care about which cell it become, so that I am very confused.

They treat Jurkats with PMA and calcium and then they differentiate into Th1 cells.

I'm pretty sure Jurkat cells are CD4 (not CD8s). Naïve T cells are either CD4 or CD8, they do not switch from one to the other (at least not that I know of).
PMA or any of the other plant based mitogens do not differentiate, they just activate. For PHA, you will need Ionomycin.
They are NOT technically naïve and as a cell line, they are limited to how much your can push them to the various t cell helper subtypes.
Depending on your goal, it might be better to use primary cells. I'm not sure you can get Jurkat cells to produce TNFa.
